The lock is not able to be manually locked and unlocked. It only works with the app now. I will attach a link to a private video showing the problem
I noticed the wyze lock does not lock and unlock via the knob anymore.

The weird (blue) substance you are referring to is called locktite. It is supposed to keep threaded screws from coming loose.
If you put the screw back in and tightened it down did the lock work properly?
Regardless, I would reach out to a Wyze support to assist you with the device.
You can call or create a ticket. You’ve documented the issue well with vignettes and could share those with the support team.
